Removed one of the datafiles from the system on purpose while the database is open. Expecting to see the error messages in the alert log. No sign of the error messages until shutdown and startup. Why is it so? I tried alter system checkpoints several time with no result whatsoever. Please help me to understand. Thanks.

Operating system is likely some Unix variant.
This is normal & expected behavior.
The file is really, really not deleted until after the last open file handle is closed.

What's the other option to detect the missing file? Will the RMAN backup be able to catch it while backing up weekly/daily?

As rman is an external program, it will not see the removed file, just like you, and so raise an error.
